I have following question:
In 6-9 Sudoku does diagonal touching also counts for 6 and 9, as it clearly does in example but not specifically mentioned in rules ?

6 and 9 may touch both by side or diagonally.
In the example it's coincidence that all 6 and 9 have some diagonal touching.
In other words, if you have a 6, you must have at least one 9 in the 8 neighboring cells and vice versa.

Twin Region Sudoku

Can you confirm if the regions in the "twin region sudoku" will be an unambiguously chain?
if other types of region are permitted, I would like to be sure that there is an unambiguous interpretation of a cyclic sequence.

Also, in the case of a row/column, could the sequence run the opposite direction?

Regions will be a chain.
It's up to you to find out the direction

Still regarding Twin Region Sudoku, just to be clear, will each region be a snake 1 cell wide which doesn't touch itself orthogonally? And for two identical snake regions, is the cyclic sequence allowed to run in opposite directions?

The both regions can be very clearly distinguished, you won't have any problems with this.
The direction may be the same or opposite, you'll have to find out
